Description:

Glutamate receptors are divided into 2 distinct classes: ionotropic glutamate receptors (iGluRs) and metabotropic glutamate receptors (mGluRs). The iGluRs consist of N-methyl-D-aspartate (NMDA) receptors and non-NMDA receptors. Non-NMDA receptors are further subdivided into 2 groups: alpha-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid receptors (e.g., GRIA1) and kainate receptors (e.g., GRIK1). The mGluRs are divided into 3 groups according to agonist selectivity, coupling to different effector systems, and sequence homology. Group I includes mGluR1 and mGluR5 (GRM5), which are coupled to inositol phospholipid metabolism. Group II, which includes mGluR2 (GRM2) and mGluR3 (GRM3), and group III, which includes mGluR4 (GRM4), mGluR6 (GRM6), mGluR7 (GRM7), and mGluR8 (GRM8), are negatively coupled to adenylate cyclase activity. Each mGluR possesses a large extracellular domain.
