Description:

Nestin is a 200kD, class VI intermediate filament protein that was originally identified by a monoclonal antibody. It was shown to be expressed predominantly in stem cells of the neural tube and its expression is absent from nearly all mature CNS cells. Upon terminal neural differentiation, nestin is downregulated and replaced by neurofilaments. It is functionally similar to other IF proteins but has a different amino acid sequence. Nestin has been the most extensively used marker to identify CNS stem cells in various areas of the developing nervous system and in vitro culture cells. Its transient expression has been suggested to be a major step in the neural differentiation pathway. Recently, Nestin expression has also been discovered in non-neural stem cell populations, such as pancreatic islet progenitors as well as hematopoietic progenitors.
